Line drawing

In Method C, during the processing, a line drawing of the item appears which is finally replaced with the composite image. Is it possible to access the final version of the line drawing and save it?

No, that's just a visualization of the focus stacking process. Why do you want it? I bet there's a Photoshop filter that does exactly what you want, and does it better.

I am preparing images of scorpions for scientific publication. In the past people used line drawings to provide key information. I would like to convert the picture to a line drawing and use it as an overlay. Ideally this would use a drawing with black lines on a transparent background.

I would just experiment with filters in Photoshop. Check edge detect filters. Maybe also pencil art filters.
