Shooting in near darkness

I have been photographing bioluminescent fungi in near darkness using a D810 and Zeiss 100 Makro Planar manual lens. If I used an autofocus lens can Helicon Remote step the focus from a set starting point even though it is too dark for the camera to focus?

Yes it can, autofocus is not used for automated focus bracketing.
