Lens Profiles

Hi. New to this forum. I'm evaluating the software now; mostly for macro work. I'm using a Nikon D90 and a Nikon D5100 with the 18-55 kit lens and a 50mm normal lens. My question is about the lens profile settings. It doesn't seem that I'm getting this right. After stacking I see unfocused areas. I attached a photo, as an example. 1. Open the DOF calculator and divide correction factor by 2.
2. Shoot and render a test stack.
3. Repeat the sequence until the resulting image is entirely focused.
